Sammanfattning : The DNA glycosylase MutYH is highly conserved throughout evolution, and homologs are found in most eukaryotes and prokaryotes examined. MutYH functions as a base excision repair DNA glycosylase that excises adenines misincorporated opposite 8-oxo-7,8-dihydro-2′-deoxyguanosine (8-oxodG), one of the most stable products of oxidative DNA damage. Sammanfattning : This thesis covers intracellular stress signaling through genotoxic stress, overriding of checkpoint control, as well as cellular redox status in hypoxic and oxidative stress Papers I and II: Caffeine has been shown to override cell cycle checkpoints in humans as well as in the fission yeast Schizosaccharomyces pombe. Sammanfattning : The use of animal organs and tissues may provide a solution to the severe and worsening shortage of human grafts. The aims of the present experimental studies were to provide a better understanding of the rejection mechanisms and give new clues for prevention of xenorejection, thereby hopefully contributing to the development of clinical xenotransplantation.
